not sure if it's a bug or not-

Assassin's blade quest requires you to kill 3 victims.  Upon completion it says the objective is to have the 'assassin's blade', which is in your possession, at the beginning of the quest.

Can't figure out what else needs to be done to complete the quest.

Also I've had the essence of on a plot of land disappear before my pioneer reaches the plot.  Any reason why this would happen?  The other resources, grain and materials are still available and the plot is still able to be settled, but the essence is gone.

The second thing is a UI bug... the essence is still there you can't see it. Sometimes grain and materials icons will also disappear, but if you hover over a square you can see the resources in the bottom right corner at the top of the box there.

 

This second location always seems to be correct even if the icons are not shown on the map.

you are wrong.  In this particular situation, I had casted a arcane monolith to prevent the AI from settling.

As my pioneer approached the resources grain and material remained, but the essence disappeared.  Rolling the cursor over the plot of land shows that the essence disappeared.  Settling on the plot did not bring the essence back.

Re:  The Assassin's Blade quest, when I reloaded the game and unequipped the sword and returned to the site; the quest was completed and blade transformed.  Returning to the site while blade is equipped did not transform the blade.

Do you think that summoning the arcane monolith caused the essence to disappear? Did you raze it in order to settle the city or was it just nearby to force the enemy to leave your territory?

nearby to force the opposing forces from settling first.

i tried reloading several times and got the same results.

what worked though was to raze the arcane monolith and recast it; and then my pioneers were able to beat the AI to the site and retain the essence in the land.

 

Not sure if the monolith drains essence from surrounding lands is a feature or not; but i don't think it is; since it doesn't do it to cities with essence already.
